چکیده
The interaction between gbcolgnorin and a spin-labeled cholesterol analogue has been tmestigated ~ EPR spee4recopy. In vesicles which were ¢econsti~ted by the freeze and thaw teelmique, ditcher evidenee was obtalned for a reorgamsatlon el the membcane at low protein content {protein/lipid ratio le~ than ! ,300). From the sign exchange interaction we were able to show a ~otem-indneed clustering of the steroid in fluid and tn gel state membranes, Tryptic cleavage of the complete N-termlnus of glycophorin vanishes the effect Whereas the removal el the s, alk aeal residues by nearandnidase digest had no influence on the EPR spectra. The [meraction seems to be eholes~ne spin label specific slnce i t was not ot~.~rv~l with an androstane spin-label

Orientation and mode of lipid-binding interaction of human apolipoprotein E C-terminal domain.
ApoE (apolipoprotein E) is an anti-atherogenic lipid transport protein that plays an integral role in lipoprotein metabolism and cholesterol homoeostasis. Lipid association educes critical functional features of apoE, mediating reduction in plasma and cellular cholesterol levels. The 10-kDa CT (C-terminal) domain of apoE facilitates helix-helix interactions in lipid-free state to promote apoE s...
متن کاملIs there a preferential interaction between cholesterol and tryptophan residues in membrane proteins?
Recently, several indications have been found that suggest a preferential interaction between cholesterol and tryptophan residues located near the membrane-water interface. The aim of this study was to investigate by direct methods how tryptophan and cholesterol interact with each other and what the possible consequences are for membrane organization. For this purpose, we used cholesterol-conta...

Nuclear-resonance spectra of phosphatidylcholine (PC) vesicles are broadened by spin-labeled PC. The broadening of the N-methyl proton line is proportional to spin-labeled PC concentration; the broadening by 1 molecule in 100 of spin-labeled PC is 9.7 Hz at 35 degrees C; the broadening is not due to collisions between vesicles, exchange of spin label between vesicles, or fusion of vesicles. The...
متن کاملImaging lipid domains in cell membranes: the advent of super-resolution fluorescence microscopy
The lipid bilayer of model membranes, liposomes reconstituted from cell lipids, and plasma membrane vesicles and spheres can separate into two distinct liquid phases to yield lipid domains with liquid-ordered and liquid-disordered properties. These observations are the basis of the lipid raft hypothesis that postulates the existence of cholesterol-enriched ordered-phase lipid domains in cell me...
متن کاملAllogeneic cytolysis of reconstituted membrane vesicles.
The successful use of lipid bilayer model membranes as targets for cytotoxic lymphocytes is described. Lipid vesicles were made from a mixture of dipalmitoyl lecithin, dimyristoyl lecithin, and cholesterol. Membrane proteins of LSTRA or EL4 tumor cells (as source of H-2 antigens), human eye muscle membrane proteins (as supporting proteins), and 51Cr marker were inserted into the lipid vesicles....
